Guillaume was especially famous for his role as butler Benson DuBois in the television series Benson. He played the role of the sarcastic and daring butler first in the comedy Soap, which began in 1977. For this role he received an Emmy in 1979. Because his character became so popular, Guillaume got his own Benson series that ran from 1979 to 1986. In 1985 he again won an Emmy.

In 1994 Guillaume lent his voice to Rafiki in the Disneyfilm The Lion King.
